# FORM — Field Operations Rhythm Model

> **F**ield **O**perations **R**hythm **M**odel.
> Compare **orthodox** regenerative and reprogramming practice (constitutive factors, dose-only optimization) with **LifeSong**: interventions as **timed pulses** aligned to biological oscillations, **cascade order** from Symphony/GEM, and **lotus time** t_L = 9T/2 from LENG.

**Vault template:** [FORM_PROTOCOL.md](../FORM_PROTOCOL.md). **Evidence (DOIs, PBM, OSKM, senescence):** **[BOOK.md](BOOK.md)**. **Work queue:** [BOUNTY_BOARD.md](BOUNTY_BOARD.md). **Narrative:** [README.md](README.md).

## A. The two paradigms

| Axis | Orthodox | LifeSong (ours) |
|------|----------|-----------------|
| **Philosophy** | Maximize pathway activation; timing is secondary to magnitude | **When, duration, and pattern** are the intervention; magnitude follows the score |
| **Free parameters** | Dose, vector copy number, constitutive vs inducible toggle, cell-type promoter | Pulse width per target **t_L**, inter-pulse gap, **order** along reverse cascade (TRIM28 → DNMT1 → UHRF1 → decouple) |
| **Tools** | Bulk OSKM, standard PBM “always on,” chronic drug exposure | Dox/optogenetic/mRNA **pulses**, **660/850 nm PBM** at structured frequency (see LS-H*), circadian-periodized hormesis (07) |
| **Output** | Pluripotency or partial reprogramming readouts at endpoint | Trajectory **shaped in time**: silencing restoration, mitochondrial coupling, SASP modulation **without** identity erasure |
| **Provenance** | Field consensus + safety-first IRB templates | Cross-project: **10** (GEM order), **05** (t_L, force hierarchy), **12** (coordinates), **07** (Orem/hormesis) |

## B. Head-to-head axes

| Axis | Orthodox | Ours | BOOK anchor |
|------|----------|------|-------------|
| **Senescence** | Marker panels; static states | **Ordered cascade** + lncRNA-before-p16 timing | BOOK §1; [10 README](../10_Project_DiscordIntoSymphony/README.md) |
| **Reprogramming** | Constitutive OSKM risk/benefit | **Cyclic / pulsed** partial reprogramming literature | BOOK §3 (LS-P2) |
| **Epigenetic maintenance** | Gene-centric edits | **UHRF1/DNMT1/TRIM28/SETDB1** sequence in time | BOOK §2 |
| **Light / mitochondria** | Irradiance-only dosing | **Frequency + phase** (10/40 Hz hypothesis) vs CcO | BOOK §4 |
| **Systemic stress** | Ad hoc fasting/cold | **t_L-matched** windows (circadian × hormesis) | BOOK §6; 07 Command |

---

## C. Murder board (honest)

1. **“Dose fixes everything.”** Constitutive Yamanaka trades identity for activation; timing is not a tuning knob afterthought.
2. **“One marker proves senescence.”** Without temporal ordering, SASP and off-target states are confounded (see GEM + BOOK senescence rows).
3. **“PBM is just red light.”** Ignoring pulse structure drops the intervention into “generic wellness” without falsifiable coupling to BOOK §4 mechanisms.

---

## D. What we take from orthodox

| From orthodox | Why | Into LifeSong |
|---------------|-----|----------------|
| Partial reprogramming safety literature | Establishes **duty-cycle** bounds | LS-P2 scan → BOOK §3 |
| Clinical PBM trials / reviews | Dose and skin safety envelopes | LS-H2 calibration |
| scRNA time-course datasets | Estimate **T** for t_L | LS-M1; GEO/ArrayExpress in BOOK §7 |

---

## E. Proof comparisons (tie to bounties)

| Question | Orthodox baseline | Our prediction | Metric / bounty |
|----------|-------------------|----------------|-----------------|
| Pulse vs continuous OSKM | Continuous default in many preclinical builds | Intermittent reduces dedifferentiation risk while retaining benefit | Literature table **LS-P2** |
| t_L vs arbitrary pulse | Ad hoc 24h/48h | **9T/2** window tracks measured **T** per gene | **LS-M1**, **LS-M2** |
| PBM frequency | CW or single Hz fad | 10 vs 40 Hz **matched** to target oscillation class | **LS-P3**, **LS-H*** |

---

## F–H. Cultivator survey, adoption, verdict (stub)

Grow **LS-*** rows into a regional/tool table (labs running pulsed reprogramming, clinical LED trials) as **LS-P2** and **LS-W*** close. **Priority:** 🔴 LS-P2 + LS-M1 → 🟡 LS-P3 + LS-H1 → 🟢 LS-W wet lab.

**Verdict (now):** Orthodox wins on **regulatory familiarity** and **constitutive toolkits**. LifeSong wins only if **time** is treated as a **first-class experimental factor** with citations drawn from **BOOK.md** — not chat memory.

## Figures (FORM_PROTOCOL §3)

Minimum set when publishing: (1) cascade **timeline** schematic (GEM order vs reversal), (2) **t_L** ladder across scales (from README table), (3) pulse train diagram for one factor, (4) PBM duty-cycle vs CW comparison sketch.
