## What this project is

The hypothesis that emerged from the **2026-05-04 D03-vs-rotenone divergence**:

If the hypothesis holds across cell types, the therapeutic implication is direct:

The COPI mechanism (operator's NIA aec2786 paper, Mazan-Mamczarz/Wind/**Leng**/Gorospe Sci Adv 2026) is upstream of the load itself. **That's the leverage point for post-mitotic cells.**

This project tests the hypothesis across cell types and works out the prevention-vs-release intervention design.

## Why this matters now

Cai Lei is dying of ALS. The original Cai Lei brief proposed the senolytic-timing biomarker (BT104+105+106) as the mechanism handle. The 2026-05-04 result on D03 confirmed it for fibroblasts + endothelium but the rotenone test exposed that **it does NOT directly transfer to neurons.** Continuing to pitch the senolytic-timing biomarker for ALS would be overclaiming.

The honest finding is more interesting and more useful: **neurons load and don't release**, which means the right intervention layer is COPI restoration (prevent the Golgi-fragmentation trigger that loads the recycler state) rather than senolytic timing (which clears already-committed senescent cells).

This is a **better offer to Cai Lei** than the original. It's also potentially a publishable mechanism distinction with broad therapeutic implications across post-mitotic-tissue diseases (neurodegeneration, heart failure, late-stage T2D, age-related macular degeneration, sarcopenia).

## What this project is NOT

- **Not a pivot away from the senolytic-timing biomarker for fibroblasts** — that finding stands; the EC senescence work is real and commercializable
- **Not a refutation of the recycler hypothesis** — the recycler hypothesis predicts cellular-state shift in stressed post-mitotic cells. Confirmed (Criterion D PASSED on rotenone). The refinement is to the *trajectory shape*: peak-release in proliferative-capable cells; monotone-load in post-mitotic cells.
- **Not a final mechanism claim** — testable predictions, not asserted truths. Multiple full-transcriptome neuron datasets needed before publishing.
- **Not a reason to delay Cai Lei outreach** — it's actually the right framing for the first message. Honest > overclaiming.
