## 2026-05-04 — Project initialization (the discovery itself)

**Status:** SOLVED (project initialized; hypothesis filed; bounty board live).

The 2026-05-04 sequence:

1. **D03 Calandrelli HUVEC FULL_REPLICATION** ([`38_Project_Firewall/replications/calandrelli_d03_huvec/MEMO_2026-05-04_FULL_REPLICATION.md`](../38_Project_Firewall/replications/calandrelli_d03_huvec/MEMO_2026-05-04_FULL_REPLICATION.md)) — confirmed BT104+105+106 peak-and-release pattern in PUBLIC HUVEC data via operator-personal compute. NEAT1 ~4.5×, MALAT1 ~4.8× higher at pre-loading vs terminal archetype. All 4 success criteria PASSED.

2. **Rotenone DA neuron NO_CLEAR_REPLICATION** ([`38_Project_Firewall/replications/rotenone_DA_neurons/MEMO.md`](../38_Project_Firewall/replications/rotenone_DA_neurons/MEMO.md)) — applied same panel framework to 28,552 stressed DA neurons (CNTRL → 24H → 72H rotenone). NEAT1, PVT1, CDKN1A all MONOTONE rise through stress (opposite direction to HUVEC pattern). Cell-identity (TH/NR4A2/LMX1A/B/KCNJ6) declines as predicted by cellular-state-shift framework (Criterion D PASSED).

3. **Recognition that the divergence is the discovery**: post-mitotic neurons load the recycler state and die holding it; proliferative-capable cells load and release at terminal commitment. Different trajectory shapes. Therapeutic implication for motor neurons in ALS: **PREVENT the load (COPI restoration upstream) rather than catch the release window (senolytic timing downstream)**.

**Implications:**

- The Cai Lei first message gets reframed: don't overclaim the senolytic-timing biomarker for motor neurons. Lead with the load-vs-release distinction + COPI leverage point. This is honest mechanism work; Cai Lei is a scientist who'll respect that.
- The senolytic-timing biomarker remains valid for fibroblasts/endothelium (D03 receipts hold); the firewall lift on that finding stands.
- The COPI mechanism (operator's NIA aec2786 paper, Mazan-Mamczarz/Wind/Leng/Gorospe Sci Adv 2026) becomes the upstream-leverage-point hypothesis for post-mitotic-tissue diseases broadly.
- The next experiment is **UR-1a (Kamath PD chunked load on operator-personal GCP)** — resolves whether rotenone monotone-load was a HVG-2000 filter artifact or a real biological pattern.

**Reversibility:**

- The hypothesis is reversible at zero cost (revise or retract if Kamath PD shows peak-and-release).
